DESCRIPCIÓN GENERAL DE INTERESES CIENTÍFICOS:
La homeostasis iónica es un proceso dinámico y un requisito fundamental para todos los organismos. Muchos minerales diferentes son necesarios para procesos bioquímicos esenciales, pero la acumulación excesiva de estos elementos puede resultar tóxica. Por ello, todos los organismos vivos han desarrollado sistemas eficientes para adquirir y almacenar estos elementos, así como mecanismos robustos para mantener concentraciones homeostáticas que eviten toxicidades y permitan responder a los cambios ambientales.
El potasio es un catión monovalente clave, necesario para múltiples aspectos del crecimiento y la supervivencia celular, tales como la compensación de cargas negativas de macromoléculas, el mantenimiento de la electroneutralidad, el turgor y volumen celular, la actividad enzimática, la síntesis de proteínas, y el mantenimiento del potencial de membrana y del pH intracelular. El objetivo general a largo plazo de nuestro grupo de investigación es generar nuevo conocimiento sobre la regulación de los transportadores de potasio tanto en plantas como en levaduras, que pueda aplicarse en el futuro a enfoques biotecnológicos para mejorar la tolerancia de las plantas a la sequía.
PROYECTOS ACTUALES:
Actualmente estamos llevando a cabo dos grandes líneas de investigación. Una está centrada en la ciencia básica y la generación de nuevo conocimiento, y la otra es de carácter más aplicado y se realiza en colaboración con un socio industrial. Ambas están relacionadas con el estudio de los mecanismos moleculares involucrados en la respuesta de las plantas al estrés, especialmente al estrés abiótico. Nuestros intereses científicos se pueden resumir de la siguiente manera:
Ciencia básica:
En las plantas, además de las funciones fisiológicas generales descritas anteriormente para el potasio a nivel celular, este catión también desempeña un papel clave a nivel de planta completa, ya que participa en procesos importantes como la apertura estomática, que regula la pérdida de agua por transpiración y la desecación vegetal. Los canales rectificadores hacia el interior son responsables de la entrada de potasio en las células oclusivas y son fundamentales para la apertura estomática. KAT1 y su homólogo cercano KAT2 son los principales canales rectificadores hacia el interior expresados en células oclusivas. Nuestro proyecto actual se centra en la caracterización de 14 proteínas que hemos identificado mediante un cribado de interacción proteína-proteína de tipo Split-Ubiquitina, en busca de interactores del canal de potasio KAT1 en la planta modelo Arabidopsis thaliana.
Estamos utilizando diversos enfoques bioquímicos y genéticos para confirmar estas interacciones y analizar su efecto sobre la actividad de KAT1 en plantas. La identificación de reguladores fisiológicamente relevantes de los canales de K⁺ contribuirá al diseño de estrategias que puedan impactar tanto en la tolerancia a la sequía como en la susceptibilidad a patógenos, dado que estos poros son responsables de la captación de CO₂ y la pérdida de agua por transpiración, además de constituir una vía de entrada para ciertos patógenos.
Ciencia aplicada con socios industriales:
Nuestro segundo eje principal de investigación, iniciado en 2018 en colaboración con la empresa SAKATA, se centra en el análisis de los perfiles proteómicos y metabolómicos de diferentes cultivares de brócoli con distintos grados de tolerancia a estreses abióticos (sequía y salinidad). El objetivo es identificar proteínas y/o metabolitos con acumulación diferencial que puedan utilizarse para predecir el comportamiento de cultivares no caracterizados en condiciones adversas de campo. Como enfoque complementario, aprovecharemos nuestra experiencia con el sistema modelo de la levadura Saccharomyces cerevisiae para sobreexpresar genes de un cultivar de brócoli tolerante y así identificar aquellos capaces de conferir tolerancia a la salinidad y la sequía. Con estos enfoques, esperamos proporcionar a SAKATA un conjunto de herramientas útiles tanto para la selección de cultivares como para programas de mejora genética.
También hemos desarrollado un sistema rápido y económico para la evaluación de extractos naturales con potencial uso como bioestimulantes, basado en el uso de los sistemas modelo Arabidopsis thaliana y Saccharomyces cerevisiae. Hemos utilizado este sistema para evaluar productos proporcionados por distintas empresas y diseñar nuevas formulaciones. Actualmente, en colaboración con la empresa CALDIC, estamos descifrando la base molecular de la tolerancia al estrés abiótico conferida por las nuevas formulaciones de bioestimulantes, y realizando ensayos en campo para evaluar su eficacia en distintos cultivos.
