GENERAL DESCRIPTION OF SCIENTIFIC INTERESTS:
Ion homeostasis is a dynamic process and a fundamental requirement for all organisms. Many different minerals are essential for key biochemical processes, but excessive accumulation of these elements can be toxic. Therefore, all living organisms have evolved efficient systems for acquiring and storing these elements, as well as robust mechanisms to maintain homeostatic concentrations that prevent toxicity and allow them to respond to environmental changes.
Potassium is a key monovalent cation required for multiple aspects of cellular growth and survival, such as balancing negative charges of macromolecules, maintaining electroneutrality, cell turgor and volume, enzyme activity, protein synthesis, and maintaining membrane potential and intracellular pH. The long-term general goal of our research group is to generate new knowledge on the regulation of potassium transporters in both plants and yeast, with future applications in biotechnological approaches to improve drought tolerance in crops.
CURRENT PROJECTS:
We are currently conducting two major lines of research. One focuses on basic science and the generation of new knowledge, and the other is more applied and carried out in collaboration with an industrial partner. Both are related to the study of molecular mechanisms involved in plant responses to stress, particularly abiotic stress. Our scientific interests can be summarized as follows:
Basic science:
In plants, beyond the general physiological roles described above for potassium at the cellular level, this cation also plays a key role at the whole-plant level, participating in crucial processes such as stomatal opening, which regulates water loss through transpiration and plant desiccation. Inward-rectifying channels are responsible for potassium uptake in guard cells and are essential for stomatal opening. KAT1 and its close homolog KAT2 are the main inward-rectifying channels expressed in guard cells. Our current project focuses on the characterization of 14 proteins identified through a Split-Ubiquitin protein–protein interaction screen, as potential interactors of the KAT1 potassium channel in the model plant Arabidopsis thaliana.
We are using various biochemical and genetic approaches to validate these interactions and analyze their effect on KAT1 activity in plants. The identification of physiologically relevant regulators of K⁺ channels will contribute to the design of strategies that may impact both drought tolerance and susceptibility to pathogens, given that these pores are responsible for CO₂ uptake and water loss through transpiration, as well as serving as entry points for certain pathogens.
Applied science with industrial partners:
Our second major research focus, initiated in 2018 in collaboration with the company SAKATA, centers on the analysis of proteomic and metabolomic profiles in different broccoli cultivars with varying levels of tolerance to abiotic stresses (drought and salinity). The objective is to identify proteins and/or metabolites with differential accumulation that could be used to predict the behavior of uncharacterized cultivars under adverse field conditions. As a complementary approach, we leverage our expertise with the yeast model system Saccharomyces cerevisiae to overexpress genes from a tolerant broccoli cultivar, aiming to identify those capable of conferring tolerance to salinity and drought. With these approaches, we hope to provide SAKATA with a useful toolbox for both cultivar selection and breeding programs.
We have also developed a fast and cost-effective system to evaluate natural extracts with potential use as biostimulants, based on the use of the model systems Arabidopsis thaliana and Saccharomyces cerevisiae. We have used this system to evaluate products provided by various companies and to design new formulations. Currently, in collaboration with the company CALDIC, we are uncovering the molecular basis of the abiotic stress tolerance conferred by new biostimulant formulations and conducting field trials to assess their efficacy in different crops.
